質量分析計とアイソトープを用いた放射線影響、
タンパク質定量の研究

我々のグループでは質量分析を用いた生体試料の解析を行っています。最新の高分解能質量分析計を用いることで、アイソトープを識別出来る分解能で質量を決定することが可能です。身の回りの物質には多くのアイソトープが含まれており、アイソトープ比を観察することで有機物中の元素組成を推定することも出来ます。我々はそのような質量分析計を用いてタンパク質の研究を行っています。

放射線の生体に与える影響

放射線照射した細胞中のタンパク質全体の解析であるプロテオーム解析、遺伝子変異のよらない遺伝子発現制御の関わるエピゲノム修飾変化を解析する事で放射線影響を調べています。このデータとセンター内のDNA, RNAの解析グループと共同でオミックス解析を目指しています。

放射線を用いたがん治療、診断法の開発

センターでは福島県立医科大学、理化学研究所などと共に、がん細胞を認識する分子を利用して治療・診断用の放射性物質をがん細胞のみに送達する技術(ドラッグデリバリーシステム;DDS)を開発しています。我々のグループでは、このがん細胞を認識する分子の解析・評価を、質量分析を用いておこなっています。

アイソトープを用いたタンパク質の絶対定量

天然にわずかしか存在しない安定同位体に置換したアミノ酸を用いてペプチドを合成することで化学的性質は同じで、質量だけが異なるペプチドを作成することが可能です。これを内部標準物質として生体由来の試料に混合し、高感度質量分析計を用いることでタンパク質の絶対定量が可能です。放射線によって変異または発現量が変化するタンパク質を発見した際に、その検証を行えるよう、アイソトープラベルした標準品を用いて高感度にタンパク質を定量するシステムの構築を行っています。
